I could not get a 2m Neopixels 60LED/m string to run the Adafruit example sketch strandtest on the Arduino Due. The exact same setup worked fine when controlled by an Arduino Uno, suggesting a level shifter may be required despite the comments by Adafruit staff.

Assuming you are powering the neopixels with 5v external power, yes you likely will need a level shifter. Some strips may work with 3.3v data signals, but it is not a sure thing. The older neopixels seemed more tolerant of voltage mismatches.
