Try another SD card with a copy of Raspbian and see if the keyboard works with that.

From the guide:
Cupcade requires a Raspberry Pi Model B with mounting holes. The new B+ board, and early Model Bs with no mounting holes, will NOT work.
There was a peripheral controller change on the B+ which is not compatible with the version of Raspbian used in the Cupcade image. We've not produced a new image yet because it's actually a pretty big undertaking, and the hardware and OS are moving targets (now with the Pi 2 out, which is totally WAY incompatible). It might be revisited once things have settled a bit, but there's no ETA.

You could try installing advmame, etc. from the Cupcade image over a B+ compatible Raspbian, but no guarantees, and advmame is -extremely- fussy about certain file locations and things.
