Pin Lyout on Proto-Screwshield R3

The board shows two rows of connected points of both sides of the board (see attached) with silk screen labels "5V" and GND".

Are these rows actually connected to the UNO, for example, 5V and GND pins? I'd like to use the rows for external isolated power rails.

Yes. These are connected to the +5 and GND rails on the UNO. But it would not be difficult to cut the traces leading to those pins to isolate the rails.

Cutting traces....exactly what I had thought. But didn't know if the board was 2 or 4 layers. Figured if 2 layer one could probably cut a couple traces.

It is a 2-layer board. You should not have difficulty identifying the traces from either the top of bottom side of the board.
