would like to build a clock with larger LED display (like 3"), can i source the LED from mouser and connect the appropriate pins to the backpack and it will still work? do i need to be concerned with the LEDs' current draw through the backpack?
ultimately what i want is a multi-timezone clock with six displays (including timezone abbreviation) all driven from the same arduino (or raspberry pi if i have to).

It would have to be a common cathode display. The HT16K33 driver chip can handle 200mA per character (~ 25mA per segment).
Display pinouts do vary. And a 3" display will probably not plug straight-in. So you will need to run jumpers from the display to the board.
