Hello,
I got my FLORA board last weekend and I was testing some code with it today. After a few successful uploads to FLORA, suddenly, when I tried to upload my code, I received the message "Couldn't find a Board on the selected port". The IDE is connected to the FLORA board and the green power light is on when the cable is connected. I tried different cables and quitting the application, but even when running the blink example sketch, I received the same error. I am hoping to resolve this issue as soon as possible as I had a deadline to reach and I am now unable to test any code.
Arduino: 1.8.16 (Mac OS X), Board: "Adafruit Flora"
Sketch uses 3956 bytes (13%) of program storage space. Maximum is 28672 bytes.
Global variables use 149 bytes of dynamic memory.
Couldn't find a Board on the selected port. Check that you have the correct port selected. If it is correct, try pressing the board's reset button after initiating the upload.
This report would have more information with
"Show verbose output during compilation"
option enabled in File -> Preferences.
Thank you in advance!

Re: FLORA "Couldn't find a Board on the selected port"
Did you try resetting once the upload attempt has starts, as it suggests?
Changing to a different USB port sometimes helps.

Yes, I tried using the reset button and different USB ports but I still received the same error.

Try a double-reset. I've found it works some times. Also, is there another serial connection open? That will interfere with an upload.

Re: FLORA "Couldn't find a Board on the selected port"
Hi I've tried resetting again and I also redownloaded the Arduino IDE and not when I upload the code, I'm getting this message: "avrdude: butterfly_recv(): programmer is not responding". Any idea how to solve this?

Re: FLORA "Couldn't find a Board on the selected port"
It's kind of drastic, but have you rebooted your Mac? USB can be finicky and I've seen devices get blacklisted. A reboot will reset everything and might get it working again.
Is the Flora showing up in the port menu?

Re: FLORA "Couldn't find a Board on the selected port"
I rebooted my computer but nothing changed. I can see the FLORA under the boards manager, I think the problem is with the port? I'm not certain, but I'm pretty sure the serial port should be connected to something with "usbmodem" in the name, and I only have two serial port options, none of which I think are the correct ones. Attaching a screenshot of "Tools" in Arduino IDE...

Re: FLORA "Couldn't find a Board on the selected port"
As I recall, you're right that it should be "usbmodem". If it can be seen by Arduino, it should show up in the port list with the board name in parens. Being in the boards menu just mneans that support has been installed and Arduino knows how to compile for it. A necessary first step, but it also has to show up in the ports menu. Have you tried resetting or double-resetting while Arduino is looking for it?
