Plugging Neopixels into ESP32-S2 Metro?

The best approach would be to connect the 5v supply to the 5v connection on the USB port using https://www.adafruit.com/product/4536. You'll probably need one of these as well https://www.adafruit.com/product/1351 and https://www.adafruit.com/product/368 to connect to the strip. That avoids any problem with multiple power sources and provides the benefit of the 500mA fuse.

Ordered! In that configuration, will I still want to connect the LED and board grounds?

Yes, all grounds must be connected together. The only exception to that is if you are isolating circuits somehow (inductively or optically).
