I have an OP-1 synthesizer with a 40-pin display cable and I'm trying to find a way to connect it to my HDMI e ink monitor to prevent eyestrain, which the original LED display causes.
I see Adafruit has an HDMI to 40-pin decoder, but this is the opposite of what I want.

Would that decoder also work for 40-pin to HDMI?
If not, is there any other product that would?

No.. the TFP401 takes HDMI input and generates output for a 40-pin dot-clock display.
I don't know of any devices that work the opposite way.
